Ponatinib (AP24534) is an orally active multi-targeted kinase inhibitor that potently inhibits all known resistant BCR-ABL mutations, including the T315I gatekeeper mutation. AP26113 is a next generation ALK inhibitor that overcomes mutations that confer resistance to crizotinib. A brief overview leading to the discovery of ponatinib will be described and the biological characterization of AP26113 will be reviewed.
