Recent advances in RNAi and gene modulation open the prospect of creating a new generation of engineered cell lines with enhanced vaccine manufacturing capabilities.

    Dr. Tripp, a Professor and the Georgia Research Alliance Chair in Vaccine and Therapeutic Studies, will share how his lab has performed a genome-wide RNAi screen to identify gene knockdown events that enhance poliovirus replication. Primary screen hits were subsequently validated with poliovirus strains and cell lines employed in vaccine production to ensure the observed increases in viral titers were broadly applicable to vaccine manufacturing platforms.

    Finally, the lab created stable knockout cell lines capable of enhance vaccine production. This approach of modulating host gene expression profiles to increase virus production has the potential to address global health by significantly enhancing vaccine production at reduced costs.
